Experiment: Bacterial strains and growth. Pseudomonas aeruginosa UCBPP-PA14 was grown in Synthetic Cystic Fibrosis sputum medium (SCFM), which mimics the nutritional environment of the cystic fibrosis lung and has been previously described (Palmer, 2007). Bacterial growth in liquid media was monitored by measuring the optical density at 600 nm (OD600) during growth at 37with shaking at 250 rpm,m and cells were harvested at an OD600 of 0.4 to 0.5. We performed two biological replications for each experiment. Proteomics experiments. We harvested P. aeruginosa UCBPP-PA14 under the same conditions as described above. After cell lysis with a French press, we spun down cell debris twice (10,000 rpm, 20 min, 4, then removed the supernatant (soluble lysates). The lysates were treated and analyzed as described below. To identify proteins, we used the P. aeruginosa PAO1 protein database, downloaded from PseudoCAP (http://www.pseudomonas.com).
